Syro II is a fully automated multi-channel peptide synthesizer equipped with a two arm pipetting robot for faster washing and dispensing of reagents. The standard system is configured with two reactor blocks with other reactor block options available including heating block kits and Tip reactor blocks. This is the system of choice for those requiring the highest peptide synthesis throughput.

Syro multi-channel peptide synthesizers are the perfect choice for demanding peptide synthesis applications. Digital syringe pumps enable exact dispensing of all reactants with microliter precision. Optimal mixing of the reactants is provided by a variable speed vortex mixer. This guarantees the best synthesis results in terms of yield and purity while minimizing waste and costs. The modular construction offers the greatest flexibility of reaction vessels and storage containers for amino acids and reagents. Reactor blocks with 18, 24, 48 or 96 positions and reactor vials in 0.4 mL (Tip reactor), 2 mL, 5 mL, 10 mL and 20 mL sizes are are available, enabling parallel synthesis of peptides in a scale range from 1–600 μmol in each reactor vial. The Syro II system is configured with two reactor blocks and can be configured with three or six 96 position Tip reactor blocks (optional accessory).
Syro XP software controls the instrument functions and calculates all amounts and volumes of amino acids and reagents required. Each cycle of the synthesis can receive a completely different protocol. This flexibility allows not only single or multiple couplings, but also complete freedom in the choice of coupling strategies within any synthesis cycle.
Pre-activation of the amino acids can be performed for all the synthesis cycles or just for individual ones. The amino acids can be activated in parallel or individually directly prior to their distribution.
The Tip Synthesis option is designed for applications that require large numbers of peptides in small quantities (1–5 μmol). Each Tip Synthesis Module can be used to synthesize up to 96 peptides at a time. Each peptide is synthesized in a 0.4 ml tip with PE frit where accurate dispensing of reagents is essential. The Syro I system can accommodate 1 Tip Synthesis Module, and the Syro II system can accommodate 3–6 modules, allowing up to 576 peptides to be made at the same time.
The left arm is equipped with a pipette tip that is connected to a dosing pump via a tube and a three-way valve. All of the reagents defined on the work area can be sampled by the left arm and delivered to any chosen container on the work area. Additionally, the left arm can take in a solvent from outside the work area via the three-way valve and also deliver it to any reachable container.
The right arm possesses a triple pipetting comb in which every single pipette tip is attached to a dosing pump via a tube and a three-way valve. The pumps take up washing solutions and reagents from storage containers outside of Syro through the valve and the distributor. All wash steps and simultaneous reagent distribution, for example piperidine distribution, can be carried out by the right arm.
This option is normally recommended for laboratories that are susceptible to humidity. The inert gas option includes all the gas controls, covers for the amino acid rack (or pre-activation rack if selected) and reagent bottles.
What Is included in the Inert Gas Option?
This option is designed for applications that require large numbers of peptides in small quantities (1–5 μmol). A Tip Synthesis Module can make up to 96 peptides at a time. Each peptide is synthesized in a 0.4 mL tip (with PE frit) where accurate dispensing of reagents is essential. The Syro I system can be equipped with one Tip Synthesis Module, and the Syro II system can be equipped with three modules in standard configuration, or customized with 6 modules, allowing up to 576 peptides to be made at the same time.
What Is included in the Tip Synthesis Option?
The Syro heating block kits are optional accessories for the Syro range of fully automated peptide synthesizers and enable the synthesis of multiple peptides in parallel at elevated temperatures.

PeptiDream Inc. relocated their head office in August 2017 and established its new research center in the Kawasaki City International Strategic Center. Non-standard peptidic compounds are of interest as a next-generation replacement for conventional protein pharmaceuticals. These compounds are composed of non-natural amino acids with various backbones. The Suga Laboratory at RCAST, University of Tokyo, is a leader in this field. It has developed the Flexizyme technology and Random Peptide Integrated Discovery (RaPID) synthesis technology to synthesize non-standard peptides in cell-free translation systems. In this research, the Suga Laboratory uses the Biotage Syro I automated peptide synthesizer for scaling up the chemical synthesis of non-standard peptides.

This application demonstrates the reproducible multiple peptide synthesis on a Syro parallel peptide synthesizer equipped with Syro heating blocks. Effective heat transfer combined with efficient vortex mixing make this system a powerful tool for the synthesis of multiple peptides with increased throughput capability.
Large numbers of related peptides are often required for many types of research, for example in proteomic studies of post-translational modifications. While this sometimes can be achieved by split-and-mix combinatorial synthesis, parallel synthesis of large numbers of peptides offers distinct advantages. These include that there is no need for decoding, well characterized compounds can be used, and that compounds can easily be transferred to other platforms, for example microarrays for parallel screening.
