SFAR C18 Selector
Reversed phase flash chromatography is a very effective purification technique for polar, ionizable and highly lipophilic compounds which cannot easily be separated by normal phase techniques. Biotage supplies pre-packed purification cartridges featuring high performance Biotage® Sfär C18 silica (formally known as HP-Sphere C18, or SNAP ULTRA C18) for a range of Biotage development and scale up flash purification platforms to support a range of different chemistries and scale up purification scenarios. This silica stationary phase has been used for nearly 10 years in validated cGMP processes worldwide and provides a unique mix of high loading capacity and resolution in packed cartridges.

Development and process development scale cartridges intended for use on Biotage® Selekt or our larger scale development platform Biotage® Isolera LS connect to the system via universal Luer-lock style connectors. Cartridges for our larger process development and scale up systems Biotage® Flash 75 / Flash 150 or Flash 400 systems require a compression module which is provided with the system, or available as a spare part (links open in a new window).
For over 20 years Biotage has been at the forefront of enabling flash purification technologies. Sfär C18 high performance silica for reversed phase purification significantly adds another and complimentary powerful solution when the most demanding of purification challenges presents itself.
Moving from lab scale to large scale comes with a number of additional considerations. Biotage® Isolera LS, Flash 75, 150 and 400 systems are extremely tough, economically viable scale-up purification systems that have been developed for this purpose. With the Biotage approach, scaling up flash purification methods is straightforward. Any method developed using a Biotage lab scale cartridge can be transferred to a larger cartridge using the scale-up factors. Once the development cartridge is determined, the lab scale method can be replayed, providing peak elution profiles for the expected scale up run which are suitable for further optimization if needed. Furthermore, Isolera LS comes with software that can automatically create linear gradients from TLC Rf spots; and then automatically convert those linear gradients into step gradients for application on a larger scale. Each of the larger flash systems may be configured with either ‘M’ or ‘L’ cartridge capability to further increase its flexibility and range.
