Sfär is the Swedish word for ”sphere”, and the name of our columns reflects the fact that we have made spherical silica a standard due to its higher surface area and higher loading capacity. Reliable and flexible, Biotage® Sfär columns deliver larger loading capacities, tighter elution bands and purer fractions than traditional flash columns.
Sfär columns are available in different sizes and media and with specific characteristics.
The HC in the name stands for High Capacity and it refers both to loading capacity and performance - this derives not only from spherical silica, but also from smaller particles size (20 µ). Combined, this high surface, small particle silica improves purification performance while reducing the amount of silica and solvent required for purification. This means you can choose between running your separation with a smaller column, using less silica and less solvent, or you can increase your loading capacity and obtain a higher amount of your compound at the end.
The D stands for Duo (i.e. Two), as Biotage Sfär gives you the possibility of external and internal liquid and dry loading, you can choose among six different ways to load your sample on the columns based on the sample characteristics.
The selection of a column can be based on the type of flash chromatography (normal phase, reversed phase, specialty phase) and the size of the molecules. First of all, though, you should decide whether you want to follow a traditional approach or trust the Biotage way to make your chemistry simpler, faster, greener. In one word, better.
Whether you plan to use liquid loading only on a closed column, or liquid and dry internal loading on a open column, use the filters below to find the most suitable columns for your need.
Biotage has made spherical silica a standard with Sfär columns. This means all columns offer the efficiency and reproducibility of spherical material. For standard separations that require a reliable normal phase media you can choose Biotage® Sfär Silica 60 μm.
The Duo version enables chemists to open the column, so that you can use the Biotage Samplet® or load your sample directly on the top of the stationary phase. Spherical silica is a standard for Sfär columns to offer the efficiency and reproducibility of spherical material. For standard separations that require a reliable normal phase media you can choose Biotage® Sfär Silica D Duo 60 μm.
The obvious choice when you need maximum resolving power to run the best possible separation with the smallest amount of silica and solvent or when you need to increase dramatically your loading capacity.
The column that combine the highest loading capacity with the flexibility of loading the sample in many different ways, the Biotage way.
Amino media combined with the added flexibility of an accessible columns to have many possible ways to you load your sample.
The column of choice when you want to increase efficiency and speed in your reverse phase separations while having the flexibility in the way you load your sample thanks to the accessible columns.
Biotage® Sfär Bio flash columns were developed with a small particle size (20 µm) and large pore size (300 Å) to provide increased resolution and effective separation of complex peptides with high performance flash reversed phase chromatography (HPFC).
This column has been developed for high performance flash reversed phase chromatography (HPFC), a very efficient technique to dramatically clean-up synthetic peptides. C4 is slightly more polar than C18 and is typically recommended when purifying either large (>40 amino acids) or very hydrophobic peptides.

Biotage developed and introduced pre-packed columns for flash purification in 1994. Today our broad selection of columns enables professionals to choose the solution which best suits their purification needs. Biotage® Sfär is a complete portfolio of flash columns, available in a large variety of sizes from 5 g to 350 g and in a range of media types, so you can purify milligrams to multi-grams of your valuable compounds quickly and easily.
The process of packing our columns has make huge steps since the first pre-packed column more than 20 years ago, reaching very high quality standards that make your projects move faster because the separation runs the first time. All Sfär columns come with a printed, unique identifier to enable recognition and categorization based on the application. Thanks to the unique QR code on every column, it is extremely quick and easy to load the column data into the instrument when used with Biotage® Selekt. This will immediately set the ideal conditions in terms of flow and pressure to run the column. Sfär columns can be run up to a pressure from 12 bar (Duo) to 20 bar (Closed) depending on the size, higher that many other commercially available columns.
Chemistry, by its very nature, involves the use of chemicals that can be harmful, toxic and potentially damaging to the environment. Drug discovery currently has a large and expensive environmental footprint so finding a solution is good for both the chemist and the environment. A first step to a greener purification is using a column with a higher loading capacity, so you can purify the same amount of sample on a smaller column and hence use lower flow rates and less solvent; and ultimately finish the job quicker. This will make your whole process greener, less expensive and also more efficient. The best way to experience that is with the Biotage® Sfär HC (High Capacity).
The unique design of Biotage® Sfär makes it possible to load your columns in six different ways, including three different dry loading options thanks to the patented Biotage Samplet® and DLV. For your difficult separations, when you want to utilize dry loading in the most convenient way, take advantage of the orange flower shaped cap of Biotage® Sfär D (Duo). When used with Biotage® Selekt, flash purification system, the instrument will suggest the optimal cartridge for your conditions and will help you in optimizing your separation even further.
