Detection of Buprenorphine and Norbuprenorphine in Oral Fluid

This application note describes a simple and robust mixed mode cation exchange solid phase extraction protocol for the extraction of buprenorphine and norbuprenorphine from oral fluid using EVOLUTE® EXPRESS CX 96-well plates. Excellent extract cleanliness allows quantitation of the analytes across the range 0.1 ng/mL to 100 ng/mL using LC-MS/MS.


Buprenorphine is an opioid derivative primarily used in pain management. It may be quantitated in bodily fluids to monitor use or indicate medical conditions. The drug concentration in body fluids correlate strongly with physiological reactions ranging from asymptomatic to comatose. Therefore, it is critical to know the route of administration and the level of opioid tolerance of the patient when results are interpreted.
