The low-level detection required for cannabis use combined with the complexity of hair testing makes for a challenging application. This poster aims to demonstrate simplified sample preparation workflow for low level analysis of THC and metabolites from hair, using Biotage Lysera and ISOLUTE SLE+ columns for sample preparation.

Solid matrix analysis by LC/MS or GC/MS is generally more involved due to the necessity of multiple manual steps to convert the sample into an extractable form. This poster aims to demonstrate workflow advantages for fingernail analysis; multi-sample homogenization, extraction and analysis for a range of drugs of abuse.
Part No: P189 rev 1Issued year: 2018File size: 1.05mbFile type: pdf
This poster describes an improved workflow for the analysis of
drugs of abuse from hair. Implementation of bead homogenization along with automated sample preparation allowed for simplified methodology. The direct solvent extraction approach avoids the need for pre-concentration while maintaining desired LOQs with either 200 or
400 μL of hair extract.

Sample preparation for hair analysis is often lengthy involving multiple manual labor steps from cutting, washing, homogenization/pulverization, digestion, sample extraction and analysis. This poster aims to demonstrate a streamlined sample
preparation workflow for hair analysis.
The workflow is evaluated using a suite of drugs of abuse, including THC and metabolites.

This application note describes the extraction of a panel of 18 steroid hormones from horse hair using ISOLUTE® SLE+. The simple sample preparation procedure delivers clean extracts and analyte recoveries greater than 80% with RSDs lower than 5% for most analytes. Linearity of greater than 0.999 is achieved for all analytes in the range 0.5-500 pg/mg of hair.

This application note describes a procedure for sample pre-treatment and extraction of THC and metabolites from human hair, using Biotage® Lysera for matrix pulverization of the sample prior to clean up using ISOLUTE® SLE+ supported liquid extraction.
Manual processing protocols were developed using the Biotage® PRESSURE+ 96 (plate format) or 48 (column format) positive pressure manifolds. For automated processing, protocols were developed using Biotage® ExtraheraTM.
