    Automated Affinity Purification of Adeno-Associate Viruses

    Adeno-associated virus (AAV) is a non-enveloped virus that can be engineered to deliver DNA to a target. The genome of this virus consists of a single-stranded linear DNA molecule of approximately 4.7 kb. The Adeno-associated virus is used as a vector for the delivery of various types of cargo to the cytoplasm and nucleus of various cells including muscle, vascular endothelial and neuronal cells.

    For the downstream workflow of virus production, affinity chromatography purification has been found to effectively remove impurities such as plasmid DNA and host cell RNA/DNA. That is then followed by an anion exchange polish to remove empty capsids.


    The CaptureSelectTM AAVX affinity matrix is specifically designed to address the high selectivity and capacity requirements for large-scale downstream purification of a broad range of naturally occurring and synthetic adeno-associated virus (AAV) serotypes used for gene therapy applications. CaptureSelectTM AAVX resin has demonstrated binding reactivity towards a set of AAV serotypes that includes AAV1, AAV2, AAV3, AAV4, AAV5, AAV6, AAV7, AAV8, AAV9, and AAVrh10 as well as synthetic serotypes.
