This application note describes the extraction of several peptides, ranging in size from a 7mer to a 54mer, from plasma and serum using EVOLUTE® ABN in a 96-well plate format. The presence of certain proteins and peptides in human biological matrices can be used as diagnostic markers for the onset of disease and other health problems in humans. The ability to detect and monitor lower levels of peptides can aid in clinical patient health assessments and therapeutic drug level determinations. The sample preparation methodology detailed in this application note can significantly increase sensitivity and detection of peptides from biological matrices on LC/MS/MS. This application note offers the user two different elution strategies due to the extensive combination of peptides that may need to be extracted either strategy may be suitable dependent upon which peptides are relevant.

•Extends the range of acidic, basic and neutral compounds that can be extracted using a single generic method, increasing method development productivity•Has an optimized pore structure which reduces the extraction of endogenous matrix components, and leads to cleaner extracts that give fewer matrix effects in LC-MS/MS analysis•Has optimized physical and chemical characteristics that improve reliability of SPE procedures•Has no secondary interactions, allowing the use of pure organic elution solvent to give high recoveries of very low drug concentrations

This poster outlines a new method for the extraction of a mixture of peptides from plasma using resin based SPE. Initial method optimization was performed using EVOLUTE ABN before comparison to the streamlined approach without plate conditioning and equilibration using EVOLUTE EXPRESS ABN. Presented at EBF 2012.

Naturally occurring corticosteroids are steroid hormones produced in the adrenal cortex and perform many functions within the body. Due to their widespread action
many synthetic corticosteroids based on the hydrocortisone (cortisol) structure have been produced. Some of their uses involve treatment of various forms of arthritis, dermatitis, hepatitis and ulcerative colitis. They can be administered orally, injected into veins or muscle, nasal sprays and drops or used as topical creams. Reliable
screening methods for these drugs are important as they are classed as ‘banned substances’ by many anti-doping agencies and regulated for use in meat producing animals.
Hello I'm trying to develop a quick method to clean up the protein/lipids from fruit and veg to allow me to minimise matrix suppression for the largest range of pesticides possible (acidic/neutral/basic). I was looking at the EVOLUTE ABN as being a good option but was also wondering if the ISOLUTE® SLE+ may be a better option. Any help you can offer would be greatly appreciated, QuEChERS is too manual and I have some automation for the SPE and potential filtration.
Specific advise on the particular suitability of one technique over the other in this case would require more details of the extraction for the fruit & vegetables. Initially, either suggestion would be a good option, but could be tailored to your particular requirements given more information on initial sample preparation.Browse more questions and answers >
